01 Interaction with psilocybin

ND

It lowers blood pressure and may lessen psilocybin's blood pressure rise. Lab findings suggest a theoretical effect on how the body clears psilocybin.

Detail Aprocitentan lowers blood pressure and may lessen psilocybin's brief blood pressure rise; this has not been studied with psilocybin. In lab studies, it inhibited the enzymes UGT1A1 and UGT2B7. The body removes psilocin mainly by glucuronidation (a UGT-driven process) and conversion to 4-HIAA. In lab studies, MAO-A, CYP2D6 and CYP3A4 also metabolized psilocin (Thomann 2024). So an effect on psilocin levels is possible only in theory. Separately, its half-life is long (about 41 hours). Sources [1][2]

02 Pharmacology relevant to psilocybin

3 listed (1 possible) · counted on reports
Liver injury
The label or source warns of liver injury, from raised liver enzymes to liver failure. Liver disease also affects how the body clears psilocin. Sources [1]
Lowers blood pressure or causes orthostasis
Lowers blood pressure, or causes a drop on standing (orthostasis) that can bring dizziness or fainting. Sources [1]
UGT inhibition (theoretical effect on psilocin levels)Possible
Inhibits UGT enzymes (glucuronidation), the main route by which the body clears psilocin. In theory this could raise psilocin levels or lengthen the effect.Possible: The source describes the effect as small, seen only in laboratory or animal studies, or theoretical. Sources [1]
